Successes Across Drug Discovery and Development

Genstruct's computer-aided causal modeling approach has accelerated drug discovery and development programs from early stage through late stage clinical development. Genstruct has successfully jumpstarted therapeutic programs by developing Causal System™ Models of disease that define novel therapeutic intervention points and novel targets that have been validated in vitro.

Typically once a target has been identified and a screen developed, the challenge often becomes lead selection. Genstruct has successfully applied its causal modeling approach to the definition of molecular mechanisms of action for lead compounds, aiding in the appropriate selection of leads to advance programs.

During lead optimization, it is important to understand off-target effects and mechanism-based toxicity. Genstruct has successfully applied its methodology to define off-target effects and mechanisms of toxicity for compounds in late stage development and safety assessment.

An increasingly critical part of success is the ability to appropriately define patient populations for clinical trial design. To this end, it is important to understand the various mechanisms and manifestations of disease, and define mechanistic biomarkers that can be used to stratify patients into groups likely to respond to the mechanism of therapy. It is also important to have mechanistic biomarkers to follow toxicity mechanisms that may be related to the drug therapy. Mechanistic causal models can also be used to define alternative indications for these compounds, expanding the therapeutic profile of newly developed and existing drugs.

Genstruct's computer-aided causal modeling has been used to:
– identify drug mechanisms of action
– identify drug resistance mechanisms for clinical compounds
– identify previously unimplicated, mechanistic targets
– identify mechanism-based drug toxicities
– identify distinct mechanisms of disease

Genstruct’s Therapeutic Focus

Genstruct is applying its computable causal modeling approach to the development of new therapeutics in three main areas of biology: Oncology, Metabolic Disorders and Inflammation. Each of these areas represents a great and largely unmet therapeutic need, is biologically complex and highly amenable to our integrative rather than the current reductionist approaches. Genstruct has demonstrated success in each of these therapeutic areas in conjunction with its pharmaceutical partners, and has been able to successfully accelerate programs through clinical development stumbling blocks and into clinical trials.

Genstruct has demonstrated success in advancing the therapeutic programs of our pharmaceutical partners. Our ability to adapt and take on new challenges have gained us experiences that span a number of disease areas and compound classes including anti-cancer, dyslipidemia, diabetes and inflammatory drugs. The following is our current list of major areas of expertise:

Oncology
Solid Tumors (breast, colo-rectal)
Prostate Cancer (specific focus on androgen resistance mechanisms)
Cancer Biomarkers
Cancer Drug Resistance Mechanisms

Metabolic Disorders
Type II Diabetes
Metabolic Syndrome
Dyslipidemia

Inflammation
Atherosclerosis
Vascular Inflammation

Genstruct's internal oncology program is specifically focused on building model sets that describe all major mechanisms of cancer, which when completed will represent the most advanced understanding of cancer to date, in the form of the most comprehensive collection of cancer models in the industry. Additionally, Genstruct has experience in cardiovascular disease and muscle biology.
